【摘要】:Bacteriathatexistinabiofilmarephenotypicallydifferentfromsuspendedcellsofthesamegenotype.Thestudyofbiofilminthelaboratoryrequiresprotocolsthataccountforthisdifference.Laboratorybiofilmsareengineeredingrowthreactorsdesignedtoproduceaspecificbiofilmtype.Alteringsystemparameterswillcorrespondinglyresultinachangeinthebiofilm.Thepurposeofthismethodistodirectauserinthelaboratorystudyofbiofilmsbyclearlydefiningeachsystemparameter.Thismethodwillenableapersontogrow,sample,andanalyzealaboratorybiofilm.1.1ThistestmethodisusedforgrowingarepeatablePseudomonasaeruginosabiofilminacontinuouslystirredflowreactor.Inaddition,thetestmethoddescribeshowtosampleandanalyzebiofilmforviablecells.1.2Inthistestmethod,biofilmpopulationdensityisrecordedaslogcolonyformingunitspersurfacearea.1.3Basicmicrobiologytrainingisrequiredtoperformthistestmethod.Thisstandarddoesnotclaimtoaddressallofthesafetyproblemsassociatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetypracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
